The Expression of NOS (Nitric Oxide Synthase) in Vagina of Pre- and Postmenopausal Women

Abstract
Objectives: To identify the expression an topographic localization of NOS in vagina of pre- and postmenopausal women and determine the change of NOS after menopause

Methods: The vaginal tissues were obtained during anterior or posterior colporrhaphy from 13 premenopausal and 10 postmenopausal women. The expression of neuronal and endothelial NOS were examined with Western blotting. The cellular localization of nNOS and eNOS was confirmed with immunohistochemistry. Trichrome staining was performed to determine smooth muscle/collagen ratios.

Results: Using immunohistochemistry, eNOS was localized in vascular endothelium and smooth muscle cells of mural and extramural arteries and arterioles. The immunoreactive pattern for neuronal NOS was found in nerve fibers, ganglias and smooth muscle fibers within nerve plexus of vaginal wall. By Western immunoblot analysis, neuronal NOS and eNOS protein expression were demonstrated. There were no differences of eNOS and nNOS expression between pre- and postmenopausal vagina tissues. By Masson¡¯s trichrome staining, increased amount of collagen deposition with vaginal atrophy was confirmed in the postmenopausal vaginal tissue.

Conclusion: Immunohistochemical and Western blotting study have demonstrated the presence of NOS in the human vagina. However, the alteration of eNOS and nNOS expression of vagina after menopause was not demonstrated.
